Cell line isolation information

The cell line HL-60/MX2 is a mitoxantrone resistant (39 nM mitoxantrone) derivative of the HL-60 cell line. HL-60 cells were established from the peripheral blood of a 35-year-old woman with acute myeloid leukemia (AML FAB M2) in 1976. The HL-60/MX2 cells exhibit a reciprocal translocation [rcpt(1;3)-(q21;p23)] not found in the parental HL-60 cell line.

Recommended propagation method

Culture medium: RPMI 1640 + 10% FBSS
Culture conditions: Humidified incubator, 95% air + 5% CO2, 37°C.
Split ratio: 1:2 to 1:4
Split frequency:
Harvesting time: when the cell density is 1.5 x 106 cells/ml
Split Method: Dilution
Seeding density: 1 – 2 x 105 cells/ml

Recommended Cryopreservation Method

Freeze medium:
Complete culture medium + 20% FBS *  + 10% DMSO (* One can increase the percentage of FBS upto 50%)
Complete growth medium supplemented with 5% (v/v) DMSO (atcc)
