T-vectors are suitable for cloning of DNA fragments that have been amplified by non-proofreading DNA polymerases. These vectors are linear DNA molecules that have thymine (T)-overhang at 3’-end of both strands. Due to 3’-overhang, self ligation of vectors is not permitted, which leads to high cloning efficiency of DNA fragments that have adenine (A) overhang at its 3’-end of both strands.
T-vectors either can be generated by several methods. They can also be purchased from several commercial suppliers.
