  • The pSilencer™ 2.1-U6 Hygro is a plasmid vector specifically designed for RNA interference (RNAi) applications in mammalian cells. 
  • It facilitates the expression of short hairpin RNA (shRNA) under the control of the U6 RNA polymerase III promoter, enabling efficient and stable gene silencing. 
  • It is widely used to generate stable knockdown cell lines.
  • It is commercially available from Thermo Fisher Scientific (Ambion™). 
  • It includes a hygromycin resistance gene, which allows for the selection of stable transfectants in mammalian cells, ensuring the long-term maintenance of gene silencing effects. 
  • The vector carries an ampicillin resistance gene, which facilitates bacterial selection during plasmid propagation in E. coli.
  • A multiple cloning site (MCS) is present in the vector, allowing for the easy insertion of target-specific shRNA sequences. For sequencing and verification of the cloned fragments, the T7 and 2.0rev primers are used. 
  • The ColE1 origin of replication (ColE1 ori) serves as the starting point for plasmid DNA replication in bacterial host cells, primarily in E. coli, ensuring high-copy plasmid amplification for further applications.
  • This vector is optimized for both transient and stable RNAi experiments, offering high efficiency and reliable gene silencing. 
  • It is suitable for a wide range of applications, including gene function studies, therapeutic research, and the development of stable RNAi cell lines.
